简介:AbstractObjective:The aim of this work was to explore the feasibility of in vivo and non-invasive monitoring of deuterium/hydrogen (2H/1H) exchange at the metabolic level upon exposure to heavy water (2H2O).Methods:The healthy female mice were randomly assigned to two groups after day 0 when both mice received standard drinking water. The treated mouse was fed with 2H2O (80%, v/v) and the control mouse fed with standard drinking water (H2O) over next 13 days. Real-time mass spectrometric analysis of volatile metabolism emitted through breathing and the skin was performed on days 1, 2, 3, 10, 12, and 13. Animal experiment was approved by the Laboratory Animal Ethics Committee of Jinan University (approval No. 20161117163322) on October 29, 2021.Results:We observed a replacement of 1H by 2H in 52 mass spectral features (60 2H/1H isotopologue pairs) for the mouse fed with 2H2O, but not for the control mouse. These included pyruvic acid and lactic acid, lysine and methyl-lysine as well as short-chain fatty acids comprising acetic acid, propionic acid, butyric acid and valeric acid.Conclusion:Secondary electrospray ionization-high resolution mass spectrometry allows monitoring in vivo2H-incorporation of metabolites in a non-invasive and real-time setup and opens new opportunities to use 2H tracing to extend current metabolic studies, especially those with a focus on anaerobic glycolysis, lysine methylation and gut microbiome via monitoring of short-chain fatty acids.

简介:摘要目的在小鼠中筛选和确定尼帕病毒(Nipah virus,NiV)融合糖蛋白(fusion glycoprotein,F)和附着糖蛋白(attachment glycoprotein,G)特异性的H-2d限制性T细胞表位。方法本研究基于NiV F和G蛋白全序列设计合成多肽库(单肽长为15个氨基酸,前后肽重复10个氨基酸),使用表达NiV F和G蛋白的DNA疫苗肌肉注射免疫BALB/c小鼠,通过矩阵设计将F和G抗原全序列肽库分别混合成肽池,取免疫后小鼠脾细胞进行IFN-γ ELISPOT检测,确定肽库中特异反应的优势表位肽。结果F肽库筛选到12条特异性优势肽,第56条多肽产生的反应最强;G肽库筛选到4条特异性优势肽,第72条多肽产生的反应最强。结论本研究使用IFN-γ ELISPOT方法筛选和确定了12条NiV F抗原特异性和4条NiV G抗原特异性H-2d限制性优势T细胞表位,为NiV免疫学与疫苗研究提供了参考。
简介:摘要:目的:分析"H2H"营养管理模式对鼻咽癌放疗患者临床结局的影响。方法:选取我院鼻咽癌放疗患者共83例,按照电脑乱序分组的方法将其分为实验A组与实验B组,其中实验A组41例,实验B组42例。传统的护理方法应用于实验A组患者,实验B组患者则在传统护理的基础上实施“H2H”营养管理模式。结果:相较于实验A组,实验B组患者口腔黏膜急性放射性反应与放疗后的缓解状况更优,生活质量评分更高,P<0.05。结论:对鼻咽癌放疗患者实施传统护理与“H2H”营养管理模式联合的护理方式能够增强患者的营养支持,改善其口腔粘膜急性放射性反应状况,提高治疗有效率,优化生活质量,安全性高,值得推广。
简介:摘要目的探讨地市级三甲医院脓毒性休克患者3 h和6 h集束化治疗完成率对患者预后的影响,并筛查影响患者预后的危险因素。方法采用回顾性分析研究方法,选择2020年1月1日至2021年12月31日在山东省聊城市人民医院重症医学科重症监护病房(ICU)住院治疗的脓毒性休克患者为研究对象,收集患者的性别、年龄、急性生理学与慢性健康状况评分Ⅱ(APACHEⅡ)、序贯器官衰竭评分(SOFA)、感染部位、致病菌、3 h和6 h集束化治疗完成情况、28 d预后等指标。采用Logistic回归分析筛选影响患者28 d死亡的独立危险因素。结果① 159例脓毒性休克患者中,28 d存活93例,死亡66例。存活组与死亡组的性别、年龄比较差异均无统计学意义。与存活组比较,死亡组APACHEⅡ评分和SOFA评分均明显升高〔APACHEⅡ评分(分):26.85±5.04比20.67±4.29,SOFA评分(分):12.86±3.02比9.37±2.51,均P<0.05〕。② 159例患者的感染部位中,腹腔感染47例(29.6%)、血流感染36例(22.6%)、肺部感染31例(19.5%)、软组织感染16例(10.1%)、泌尿系感染13例(8.2%)、胆道系统感染12例(7.5%)、其他部位感染4例(2.5%);128例检出病原菌,检出率为80.5%,其中革兰阴性(G-)杆菌90例(56.6%)、革兰阳性(G+)球菌27例(17.0%)、真菌11例(6.9%),前3位致病菌分别为大肠埃希菌49例(30.8%)、肺炎克雷伯菌21例(13.2%)、金黄色葡萄球菌15例(9.4%),差异均无统计学意义。③ 159例患者中101例(63.5%)完成3 h集束化治疗,其中存活组67例(72.0%)、死亡组34例(51.5%);106例(66.7%)完成6 h集束化治疗,其中存活组70例(75.3%),死亡组36例(54.5%),两组间比较差异均有统计学意义(均P<0.05)。④将单因素分析中影响预后的因素(APACHEⅡ评分、SOFA评分、3 h和6 h集束化治疗完成率)纳入二元Logistic回归分析,结果显示,APACHEⅡ评分、SOFA评分、3 h集束化治疗完成率是影响患者28 d死亡的独立危险因素〔优势比(OR)分别为1.216、1.303、0.402,均P<0.05〕。结论脓毒性休克患者APACHEⅡ评分及SOFA评分越高,病情越重,预后越差,提高3 h及6 h集束化治疗完成率,尤其是3 h集束化治疗完成率,可降低患者病死率,改善预后。
简介:摘要目的分析2014—2021年我国禽流感相关环境中低致病性H3、H4和H6亚型禽流感病毒的特征。方法在31个省、自治区和直辖市设立监测点,开展禽流感相关环境样本的采集、甲型流感病毒核酸检测、病毒分离、深度测序和致病性相关分子位点的分析,确定其中常见的H3、H4和H6亚型禽流感病毒在不同地区、不同场所和不同样本类型中的分布及病毒变异特征。结果共采集388 645份样本。低致病性H3(0.56‰)和H6亚型(0.53‰)的分离阳性率高于H4亚型(0.09‰);H4亚型病毒在活禽市场的分离阳性率高于其他场所,差异有统计学意义;H3和H6亚型病毒在污水样本的分离阳性率高于其他样本类型,差异有统计学意义;H3、H4和H6亚型病毒南方地区分离阳性率高于北方地区,差异有统计学意义;12月份是病毒最为活跃的时间。致病性相关分子位点的分析显示,H3、H4和H6亚型病毒结合禽流感病毒受体,与致病性增强相关的某些基因位点发生了突变。结论低致病性禽流感H3、H4和H6亚型病毒在活禽市场和污水中具有较高的分离阳性率,3种亚型病毒的分布具有明显的地域特征和季节性,基因特征仍表现为低致病性禽流感特点。
简介:AbstractObjective:To analyze the proportion of peripheral regulatory T cells (Tregs) and the expression of the immune checkpoint molecules T-cell immunoglobulin and ITIM domain (TIGIT) and CD226 on Tregs in patients with recurrent spontaneous abortion (RSA).Methods:The proportion of CD3+CD4+CD25+Foxp3+ Tregs and the expression levels of CD226 and TIGIT on Tregs in 30 normal pregnant women and 28 patients with RSA were determined via flow cytometry.Results:The proportion of Tregs in the RSA group (4.41 % ± 1.54%) was significantly lower than that in the control group (5.27% ± 1.52%, P = 0.0374). Compared with the normal pregnant women, patients with RSA showed decreased TIGIT expression (54.75 ± 9.70% vs. 63.07 ± 12.48%, P = 0.0066) and increased CD226 expression on Tregs (25.59% ± 8.22% vs. 20.46% ± 6.97%, P = 0.0168). The ratio of CD226 to TIGIT in the RSA group (0.48 ± 0.19) was higher than that in the control group (0.34 ± 0.15, P = 0.0027). The proportion of TIGIT+CD226+ Tregs was significantly lower in patients with RSA (9.30% ± 4.95% vs. 13.43% ± 4.72%, P = 0.0020) than in the controls.Conclusions:Patients with RSA show a reduced proportion of Tregs and an imbalance in the expression of TIGIT and CD226 on Tregs.

简介:【摘要】目的:分析三碘甲状腺原氨酸(T3)、甲状腺激素(T4)与促甲状腺激素(TSH)水平联合检验甲状腺疾病的诊断价值。方法:共纳入甲状腺疾病患者96例,其中甲状腺功能亢进患者50例(甲亢组)、甲状腺功能衰减患者48例(甲减组);以同时期的48例健康体检者为对照组,时间为2020年11月-2021年9月,比较三组T3、T4与TSH水平。结果:甲亢组T3与T4水平高于对照组,TSH水平低于对照组;而甲减组T3与T4水平低于对照组,TSH水平高于对照组,差异有统计学意义(P<0.05)。结论:T3、T4与TSH指标水平检验可为甲亢及甲减疾病鉴别提供可靠参考,值得推广。
简介:[摘要]嵌合抗原受体T(CAR-T)细胞治疗被认为是治疗复发或难治性肿瘤,特别是血液系统恶性肿瘤的有效方法。尽管最初批准的抗CD19 CAR-T疗法取得了令人印象深刻的结果,但也经历了高复发率和耐药性等挫折,促使人们需要发现更有效的工程CAR-T细胞用于治疗。CAR-T细胞在结构和制造方面的创新导致了疗效和持久性的显著改善,特别是随着第四代CAR-T细胞的发展。与免疫调节剂配对,第四代和下一代CAR-T细胞的使用将不会因为细胞毒效应而受到限制,将成为克服肿瘤微环境的有效工具。本文综述了CAR-T细胞结构在胞外区、跨膜区和胞内区的最新变化,结合创新的制造技术和改进的细胞来源,展望了CAR-T细胞治疗的未来发展前景。
简介:【摘要】目的 骨关节炎(Osteoarthritis,OA)是一种累及关节软骨及非软骨组织的骨关节高发病,具有症状进行性加重的特点,严重影响患者生活质量,早期诊断及治疗成为临床工作亟待解决的问题。T2 mapping作为一项磁共振的新技术,可以针对损伤组织中各成分含量的改变进行定量分析,从而检测出早期OA的软骨变性。三维梯度回波T2*加权脂肪抑脂(three-dimensional Gradient recalled echo T2*weighted imaging fat suppression,3D GRE T2*WI fs)是在GRE T2*WI基础上研发的扫描序列,扫描时间较常规PD抑脂像节省了65秒的时间,可大范围、无间隔连续薄层扫描。本研究旨在评估T2 mapping及3D GRE T2*WI抑脂像在髋关节OA诊断中的应用价值及其临床意义。
简介:AbstractAvian influenza remains a threat to human wellbeing. Hypochlorite derivatives are commonly used as disinfectants to prevent the spread of the disease. The World Health Organization (WHO) has listed chlorine dioxide (ClO2) as an A1-level, safe, and efficient disinfectant. In this study, we tested the efficacy of ClO2, in aqueous solution and gas forms, against avian influenza A (H7N9) virus. The virus suspension was mixed with ClO2 aqueous solutions of various concentrations and for various time intervals. Aliquots of the mixture were then serially diluted, and the 50% tissue culture infective dose (TCID50) was measured with a hemagglutination test on MDCK cells. ClO2 gas produced from generators was introduced in a chamber containing the virus suspension in a Petri dish. The infective activity of the surviving virus was measured by the hemagglutination test. An aqueous solution of ClO2 at 126 µg/mL for 15 s was effective given that no surviving virus was detected with the hemagglutination test. ClO2 gas at >5 µL/L sustained for 1 h inactivated the virus effectively, while at 2.5 µL/L for 1 h, it only partially inactivated the virus. ClO2 as gas or aqueous solution at a certain concentration is effective in inactivating the H7N9 virus, and can be applied for the decontamination and disinfection of environments.
