简介:Hepatocellularcarcinoma(HCC)isoneofthemostdeadlyhumancancers,butitisverydifficulttoestablishananimalmodelbyusingsurgicalspecimens.Inthepresentexperiment,histologicallyintactfreshsurgicalspecimensofHCCweresubcutaneouslytransplantedinnon-obesediabetic/severecombinedimmunodeficienccy(NOD/SCID)mice.Thebiologicalcharacteristicsoftheoriginalandthecorrespondingtransplantedtumorsandcelllineswereinvestigated.Theresultsshowedthat5newanimalmodelsand2primarycelllinesweresuccessfullyestablishedfromsurgicalspecimens.Hematoxylin-eosinstainingshowedthatxenograftsretainedmajorhistologicalfeaturesoftheoriginalsurgicalspecimens.Thetwonewcelllineshadbeencultivatedfor3yearsandsuccessivelypassagedformorethan100passagesinvitro.Themorphologicalcharacteristicsandbiologicfeaturesofthetwocelllinesweregeneticallysimilartotheoriginaltumor.Thesubcutaneoustransplantanimalmodelswithhistologicallyintacttumortissueandprimarycelllinescouldbeusefulforinvivoandinvitrotestingofanti-cancerdrugsandbeidealmodelstostudyvariousbiologicfeaturesofHCC.
简介:ObjectiveTostudyexpressionofadenoviral-mediatedHath1-EGFPgeneintheguineapigcochleaaftertransferthroughintactroundwindowmembrane(RWM),andtoassessitseffectsonhearing.MethodsTwentyadultguineapigswereused,ofwhich:12weresurgicallyinoculatedwithAd-Hath1-EGFPinthebonygrooveofroundwindowniche,and8withartificialperilymph.Auditorybrainstemresponse(ABR)thresholdsweredeterminedinallanimalsbeforeand5daysaftersurgery.Onpost-surgeryday5andday14,animalsweresacrificedandwholemountsofcochleaandfrozensectionswereexamined.ResultsABRtestsshowednosignificantchangeofhearingafterthesurgery.StrongfluorescencestaininginthecochleaewasseeninAd-Hath1-EGFPgroups.Thehighestlevelsofgeneexpressionwereseeninthepost-surgeryday5groupwithlittledecreaseonpost-surgeryday14.Thecontralateralcochleaandthoseinthecontrolgroupswerefreeoffluorescencestaining.ConclusionThetransgenicHath1-EGFPcanbeeffectivelydeliveredintotheinnerearthroughintactRWM,inanatraumaticmanner.
简介:Aim:Toassessthequalityofhigh-resolutionCTsectionplanes(HRCT),multi-planarreformation(MPR)and3-dimensionalvolumerenderedcomputertomography(3D-CTVR)werehereusedinthefinedifferentialdiagnosisofossicularchaininthecaseofconductivehearinglosswithintacttympanicmembrane.Methods:Here,17casesofotosclerosisand22casesofossicularchaindeformitywereselected.Allpatientshadnormalexternalearcanals,intacttympanicmembranes,conductivehearingloss,typeAtympanograms,andnegativeGelle'stests.Therespectiveradiologicalreportsofthestatusoftheossiclesvia3protocolswerecomparedtosurgicalfindings.Thequantitativeassessmentsoftherepresentationofdifferentsegmentsoftheossicularchainwerebasedona3-pointscoringsystem.Results:MPRandCTVRimagingbothshowedtheintegrityofwholeossicularchainwell.MPRandCTVRimagingwerefoundtobesuperiortosectionplaneswithrespecttoshowingthesuperstructureofthestapesandmalformations(P>0.05).Conclusion:CTVRandMPRimagingwerefoundtobebetterabletoshowthewholeossicularchainintheconductivehearinglosswithnormaltympanicmembranes.Furthermore,theuseofthesetechniquescanhaveprofoundcontributivevalueinthedifferentialdiagnosisofotosclerosisandossicularchainabsenceormalformation.
